学术成就
上皮细胞定位于机体体表与内腔表层,在维持机体稳态平衡中发挥着重要的作用,其功能紊乱可导致多系统疾病的发生。张艺林博士以上皮细胞为研究对象,重点围绕上皮细胞的免疫调控、离子转运及细胞间对话等生理功能展开研究工作,以探究上皮细胞及其离子信号网络在人类健康与疾病中的关键调控作用机制,为靶向调节上皮细胞功能的疾病干预策略提供新的科学依据。
主要创新点
1)发现多种病原体感染可诱导气道上皮细胞内氯离子稳态失衡,推动生理性炎症向病理性炎症转化。在此过程中,首次发现一条全新的氯离子-SGK1信号通路,并系统性地阐述了该通路介导支气管扩张、新冠肺炎、肺弓形虫病等呼吸系统疾病炎症迁延不愈的病理新机制,以及靶向抑制该通路的抗炎治疗新策略。
2)揭示了慢性气道炎症性疾病发生时,气道上皮细胞释放的多种促炎细胞因子可作用于邻近的气道平滑肌细胞,诱导其过度收缩;验证了靶向调节信号下游的CaV1.2钙离子通道具有缓解模型小鼠气道高反应性的作用,提示该策略可作为哮喘等呼吸系统疾病潜在的治疗方案。
3)阐明了生殖道上皮细胞通过激活离子通道调节生殖道管腔离子微环境形成的机制,并揭示了这一特殊的离子微环境在宿主防御与生殖中的重要作用。
